CB1- and CB2-receptors have, so far, been found in various types of cells that can be found in human skin, e.g. in:
- Mast cells
- Nerve fibers
- Sweat glands
- Hair follicles-Epithelium
CBD – Usage potential with Psoriasis?
Psoriasis is a widespread chronic- inflammatory skin disease/ Autoimmune disease, that cannot yet be healed and in cases of many patients, cannot be treated
Psoriasis is characterized by a hyperproliferation of keratinocytes and overproduction and the infiltration of inflammatory mediators into the skin
In principle, the following applies:
- Skin cells have Cannabinoid receptors
- The endocannabinoid anandamide inhibits the differentiation of keratinocytes
- Cannabinoids act against other autoimmune-like rheumatoid arthritis or Crohn’s disease, by influencing the expression of cytokines
- Cannabinoids inhibit the multiplication of certain other rapidly proliferating human cell lines – e.g. various tumor cells – noticeably
- CBD has strong anti-inflammatory traits
- CBD is lipophilic and permeates deep into the skin. Also, CBD is very compatible and causes no psychotropic side effects
A lot of things talk in favor of therapeutical potential of usage of CBD for treatment of skin diseases.
In-vitro-Study: CBD inhibits the multiplication of keratinocytes
4 natural cannabinoids are isolated from hemp: THC1, CBD2, CBN3 und CBG4.
Special cell lines of human keratinocytes are treated and cultivated with each of these cannabinoids in various concentrations.
After a 72-hour Incubation period, the cell numbers are determined.
All of the four tested Phytocannabinoids inhibit the proliferation of keratinocytes in vitro.
Cannabidiol inhibits the proliferation, in comparison with the tested substances, the most.
A 50% inhibition of the cell growth is achieved with a 2 µM („IC50“) concentration of cannabidiol.
The maximum, near complete inhibition of growth, is achieved with < 10 µM CBD.
CBD acts anti-inflammatory
The tests conducted on animals (a murine model of rheumatoid arthritis) acts transdermally with the help of Ethosomes1, the applied Cannabidiol acts anti-inflammatory.
Numerous publications verify the anti-inflammatory Potential of Cannabidiol in vitro – e.g. on cell cultures – or in vivo in tests conducted on animals.
Depending on the concentration, Cannabidiol inhibits the distribution of inflammatory cytokines (Interleukins IL- 1beta, IL-6, IL-17) and enhances the production of anti-inflammatory cytokines (Interleukin 10) in cells and intervenes into processes that are disrupted during autoimmune diseases.
CBD – Usage potential with acne?
- Acne is marked by an excessive production of sebum in the tallow glands in relation to inflammatory changes in the skin and local bacterial infections of the affected tissue.
- Just like keratinocytes, so do sebocytes function as cannabinoid receptors – especially CB2 – on their surface
- Through sebocytes, endocannabinoids enhance in vitro the sebum production in cell lines, dramatically
In-vitro-Study: CBD regulates the excessive tallow production of human sebocytes
Special cell cultures of human sebocytes (tallow glands cells) are partially treated and cultivated with substances which increase tallow production (AEA1among others!)
After an incubation period of 24 hours, the extent of the lipid synthesis is determined.
A lipid synthesis of sebocytes, excessively enhanced by AEA, is reduced with CBD (depending on concentration). 10 µM CBD is enough to normalize a lipid production which was enhanced 2.3 times.
It is examined which lipid classes produce stimulated1 Sebocytes in particular, and how this lipid pattern is influenced by the regulating influence of CBD.
10 µM of CBD, in the stimulated sebocyte culture, is enough to completely normalize the excessive production of all lipid classes.
(Single exception: Diglycerides (DG); Production is also significantly reduced.)
In-vitro-Study: CBD has an antiproliferative effect on sebocytes in the preservation of health
The influence of CBD on health and proliferation of the cultivated Sebocytes was examined.
Incubation over a period of 24-72 hours with 1–10 µM CBD reduced the (in case of acne, undesirable) sebocyte proliferation without toxic effects.
Longer incubation with 10 µM or manifold higher concentrations of CBD (50 µM) also impairs the health of cell cultures (in vitro).
In-vitro-Study: CBD acts anti-inflammatory
The sebocyte cultures are, with the use of lipopolysaccharide (LPS), provoked into an inflammatory reaction and the effect of varying CBD- concentrations on the extent of these reactions are measured.
10 µM of CBD is enough to greatly inhibit the excessive production of typical inflammation parameters (IL-1B, IL-6, TNFα) in the LPS-provoked sebocyte culture.
This finding is backed by comparable results after an acne-stimulating pretreatment of the cell cultures.
In cell culture models, on sebocytes (in vitro) as well as in tallow gland model in the tissue (ex vivo), cannabidiol has the following effects:
- antiproliferative in the preservation of health and
and therefore cover three active principles of an effective anti-acne therapy.
Note: An early, rough estimate allows the scale of the needed CBD-concentration for the inhibition of the sebocyte proliferation, the excessive tallow production caused by sebocytes and the inflammation processes to appear achievable (slide 6)!
